雌激素受体α在睾酮影响人脐静脉内皮细胞衰老中的重要作用

摘    要:目的观察睾酮对过氧化氢(H2O2)诱导衰老的人脐静脉内皮细胞(HUVECs)的影响,并初步探讨其作用机制。方法将内皮细胞分为正常对照组、H2O2对照组、不同浓度睾酮组(3×10-9~3×10-6mol/L)、ICI182,780组和氟他胺组。用SA-β-半乳糖苷酶细胞化学检测法观察各组细胞衰老情况,Western blot法检测细胞中雌激素受体α(ERα)水平。结果与正常对照组比较,H2O2对照组可以引起HUVECs衰老细胞明显增加。3×10-9mol/L、3×10-8mol/L、3×10-7mol/L睾酮组均具有延缓H2O2诱导HUVECs衰老的趋势,3×10-6mol/L睾酮组却具有相反的作用。3×10-9~3×10-6mol/L睾酮组ERα各条带吸光度值分别为113.54±8.09、165.69±7.09、143.63±6.84、80.81±5.29。结论睾酮对H2O2诱导HUVECs衰老的影响与其剂量有关,而且睾酮还呈剂量相关性调节H2O2诱导HUVECs的ERα表达。

Abstract:Objective To investigate the effects of testostrone on the senescence of human umbilical vein endothelial cells(HUVECs)induced by hydrogen peroxide(H2O2),and preliminarily investigate the mechanism of it.Methods The endothelial cells were divided into normal control group,H2O2 control group,testosterone at different concentrations groups(3×10-9-3×10-6 mol/L),ICI182,780 group and flutamide group.The percentage of senescent cells in each group was studied by means of SA-β galactosidase staining,and the expression of estrogen receptor α(ERα)was detected by Western blot.Results H2O2 induced senescence of HUVECs significantly.Testosterone at low concentrations(3×10-9 mol/L,3×10-8mol/L and 3×10-7mol/L)had the trend of delaying the senescence of HUVECs induced by H2O2,whereas higher concentration(3×10-6 mol/L)had the opposite effect.ERα expression in the HUVECs stimulated by H2O2 could be regulated by testosterone(113.54±8.09,165.69±7.09,143.63±6.84,80.81±5.29 for testosterone at concentrations of 3×10-9 mol/L,3×10-8 mol/L,3×10-7 mol/L and 3×10-6 mol/L,respectively).Conclusions Testosterone modulates not only the senescence,but also the ERα expression of HUVECs induced by H2O2 in a dose-related manner.
