摘    要:目的检测吡喹酮以及苯并咪唑类药物对重组的华支睾吸虫乳酸脱氢酶(CsLDH)酶促功能的影响,探讨其药物作用机制。方法将不同浓度的吡喹酮、阿苯达唑、芬苯达唑和甲苯咪唑加入CsLDH所催化的丙酮酸还原成乳酸(正反应)以及乳酸氧化成丙酮酸(逆反应)的标准反应体系当中,紫外分光光度法测定NADH在A340处吸光度,SPSS16.0统计软件分析试验数据。结果相比于对照组,9mmol/L的吡喹酮对正、逆反应的抑制均可达到94%(P<0.01);0.2 mmol/L的阿苯达唑对正、逆反应的抑制分别为84%(P<0.01)和79%(P<0.01),0.06 mmol/L的芬苯达唑对正、逆反应的抑制分别为92%(P<0.01)和86%(P<0.01),0.1 mmol/L的甲苯咪唑对正、逆反应分别为98%(P<0.01)和87%(P<0.01)。结论吡喹酮、阿苯达唑、芬苯达唑和甲苯咪唑可能以CsLDH为作用靶点杀伤华支睾吸虫而发挥治疗作用。

Effect of anti-parasite drugs on the recombinant lactate dehydrogenase from Clonorchis senensis
Abstract:To investigate the effect of anti-parasite drugs,different concentrations of praziquantel(PQZ),abendazole(ABZ),fenbendazole(FBZ) and mebendazole(MBZ) were added into the standard reaction system catalized by lactate dehydrogenase from Clonorchis sinensis(CsLDH) and the ABSorbance of NADH at 340 nm wave length was detected.Data were analyzed by using SPSS16.0 software.Compared with the controls,the inhibition rate of 0.2 mmol/L PQZ on CsLDH-catalyzing oxidation-reduction reaction was 94%(P<0.01),while those on CsLDH catalyzing oxidation reaction of 0.2 mmol/L ABZ,0.6 mmol/L FBZ and 0.1 mmol/L MBZ were 84%,92% and 98% respectively(P<0.01)and those on CsLDN reduction reaction were 79%,86% and 87% respectively(P<0.01),From these observations,it suggests that CsLDH may be the target of drug-effect against C.senensis.
