血管活性肠肽对AECOPD患者PBMC中NF-κB表达的影响

摘    要:目的 观察核因子κB(NF-κB)与慢性阻塞性肺疾病(COPD)炎症反应的关系,探讨血管活性肠肽(VIP)对慢性阻塞性肺疾病急性加重期(AECOPD)患者外周血单个核细胞(PBMC)中NF-κB的影响.方法 采集15名正常人和15例AECOPD患者的PBMC,来自正常人的PBMC为正常对照组,来自AECOPD患者的PBMC分成两组,分别为AECOOPD对照组和VIP组,采用免疫细胞化学方法检测三组间细胞核中NF-κB蛋白表达量的差异.结果 ①AECOPD对照组NF-κB阳性细胞表达量明显高于正常对照组(P<0.05);②AECOPD对照组细胞核中NF-κB蛋白表达量明显高于VIP组(P<0.05).结论 AECOPD患者的PBMC中NF-κB的表达量较正常健康人高,表明NF-κB参与了AECOPD炎症反应过程;应用VIP可降低AECOPD患者PBMC中NF-κB的表达量,推测VIP对AECOPD具有一定的抗炎作用.

Abstract:Objective To explore the expression of NF-κB in peripheral blood mononuclear cells (PBMC) from acute exacerbation of chronic obstructive pulmonary disease(AECOPD) patients and the inhibitory effect of vasoactive intestinal peptide (VIP). Methods PBMC isolated from 15 healthy subjects and 15 AECOPD patients were cocultured. PBMC of healthy subjects were acted as normal group. PBMC of AECOPD patients were devided into two groups, AECOPD group and VIP group where VIP were added. The expression of NF-κB was determined by immunocytochemical staining. ResultsThe percentage of NF-κB nucleus positive cells was higher in AECOPD patients than in normal controls( P <0.05). The percentage of NF-κB nucleus positive cells was higher in AECOPD controls than in VIP group ( P <0.05). Conclusions The results indicated that NF-κB may play a pathogenic role in the development of AECOPD and that VIP can inhibit inflammatory reaction of AECOPD.
