摘    要:目的研究不同中医治法对帕金森病大鼠氧化应激反应的影响。方法采用经典的6-羟基多巴胺损毁注射法制作PD模型,并应用活血化瘀(代表方桃红四物汤)、涤痰熄风(代表方涤痰汤)、滋阴熄风(代表方天麻钩藤饮)和复合治法(代表方复方地黄方)等不同治法的方剂进行治疗,同时设立正常对照组、假手术组作对照。测定大鼠中脑黑质纹状体部位丙二醛、谷胱甘肽含量,谷胱甘肽过氧化物酶、超氧化物歧化酶的活性。结果模型组SOD、GSH、GSH—Px下降,MDA升高。各中药组均可以改善模型组的异常变化,差异均有统计学意义(P〈0.05,或P〈0.01)。桃红四物汤组与涤痰汤组比较,各指标差异均无统计学意义(P〈0.05)。天麻钩藤饮组与桃红四物汤组比较,在SOD、MDA、GSH指标上差异均有统计学意义(P〈0.05,或P〈0.01);与涤痰汤组比较,在SOD、MDA指标上差异均有统计学意义(p〈0.05)。复方地黄方与桃红四物汤比较,在SOD、MDA和GSH—Px指标上差异均有统计学意义(P〈0.05,或P〈0.01);与涤痰汤比较,在SOD、MDA和GSH指标上,差异均有统计学意义(P〈0.01);与天麻钩藤饮比较,在GSH指标上,差异有统计学意义(P〈0.01)。结论各种治法均有明显的抗氧化应激损伤的作用,但以复合治法为好。

Influences of different TCM protocols on oxidative stress in a Parkinson's disease rat model

Abstract:Objective To investigate influences of different TCM therapies on oxidative stress in a Parkinson's disease (PD) rat model. Methods The PD rat Model was constructed by the classic 6-OHDA injection method, and treated with different TCM protocols, including Ziyin Xifeng (Tianma Gouteng Decoction), Huoxue Huayu (Taohong Siwu Decoction), Ditan Xifeng (Ditan Decoction), and a compound therapeutic method (Compound Dihuang Decoction). Changes of oxidative stress in different PD rat groups were observed and compared with the normal and sham groups. GSH and MDAs, SOD and GSH-Pxs of substantia nigra and corpus striatum in PD rats were measured. Results Abnormal changes were improved in all TCM treatment groups, and the difference was statistically significant (P〈0.05, or P〈40.01). There was no statistically significant difference in any of the parameters measured between Taohong Siwu Decoction and Ditan Xifeng Decoction groups (P〉0.05). There was significant difference in MDA, SOD and GSH-Px between Tianma Gouteng Decoction and Ta'ohong Siwu Decoction groups (P〈0.05, or P〈0.01), and in MDA and SOD between Tianma Gouteng Decoction and Ditan Decoction groups (P〈0.05, or P〈0.01). There was significant difference in MDA, SOD and GSH- Px between Compound Dihuang Decoction and Taohong Siwu Decoction groups (P〈0.05, or P〈0.01); in MDA, SOD and GSH between the Compound Dihuang Decoction and Ditan Decoction groups (P〈0.01); and in GSH between the Compound Dihuang Decoction and Tianma Gouteng Decoction groups (P〈40.01). Conclusions All the TCM therapeutic protocols are effective against oxidative stress, of which the compound TCM protocol is the most effective.
