日本血吸虫与华支睾吸虫和姜片虫之间的交叉抗原及其血清学反应性

摘    要:应用SDS-PAGE分离AWA、CSA和FSA三种抗原的组分蛋白后,再以酶联免疫印迹技术(ELIB)进一步鉴定其组分蛋白的特异性蛋白带,分别显示12条、19条和21条,其分子量(MW)范围分别为13~64kDe、16~198.5kDa和14~150kDa。AWA组分蛋白与兔抗SEA免疫血清出现3条交叉反应带.MW为17kDa、14kDa和13kDa,与抗CSA的兔血清及华支睾吸虫病人血清显示3条(38kDa、17.5kDa及17kDe)交叉反应带.与抗FSA的兔血清及姜片虫病人血清呈现5条(64kDa、59kDe、53kDe、17.5kDa和17kDa)交叉反应带。实验结果表明,日本血吸虫成虫不仅与其虫卵之间存有交叉抗原,而且与华支睾吸虫和姜片虫成虫之间也存有交叉抗原及血清学交叉反应性。本研究为今后制备特异的血吸虫病血清学诊断抗原,提供了科学依据。

Abstract:The enzmpe-linked immunoblotting technique (ELIB) respectively using AWA, CSA and FSA fractional proteins as antigens was used to detect the specific reactivities in sera from rabbits immunized with several trematode antigens, and from rabbits and patients infeCted with several trematedes. The results showed that 12, 19 and 21 specific bands were separated from AWA,CSA and FSA. The range of their molecular weights was 13-64 kDa, 16- 198. 5 kDa and 14 -150 kDa respectively.The AWA fractionated proteins recognized by sera from rabbits immunized with SEA appeared three cross reactive bands with molecular weights ranging from 13 to 17 kDa. The cross reactivities of the AWA fractional proteins appeared in sera from rabbits immunized with CSA and FSA, and patients infeCted with C. sinensis and F. buski demonstrated three and five cross reactive bands with the molecular weights ranging from 17 to 38 kDa and 17 to 64 kDa respectively.The results indicated that the cross antigen present between S. japonicum adult worms and eggs. The present study also proVed that S. japonicum shares its antigens and appears cross reactivitids with C. sinensis and F. buski and cross antigens and their cross reactivities also appear between the latter two trematodes. The present study provides the scientific basis for preparing the specific antigens for diagnosis of schistosomiasis japonica.
