Notch1信号蛋白在卵巢癌中的表达及临床意义

目的：检测Notch1在卵巢上皮性肿瘤的表达,探讨其在卵巢癌的形成和转化过程中的表达特点及其与卵巢癌临床病理学参数的关系。方法：采用免疫组化法、逆转录聚合酶链反应（RT-PCR）法及Western blot法检测109例卵巢癌（其中65例为单侧卵巢癌）、65例单侧卵巢癌患者健侧卵巢（配对对照）及48例正常卵巢组织（正常对照）中Notch1的表达。结果：（1）卵巢癌中Notch1阳性表达率95.4%（104/109）]显著高于配对对照组7.7%（5/65）]和正常对照组6.3%（3/48）]（P〈0.01）,而后两组则无显著差异（P〉0.05）。（2）卵巢癌中Notch1的表达与临床分期（FIGO）及病理分化程度有关（P〈0.01）,临床分期越高、分化程度越低,Notch1的表达越高;而与年龄、家族史、肿瘤位置、有无腹水、有无周围组织浸润、有无淋巴结转移、病理类型无关（P〉0.05）。结论：Notch1在卵巢癌的发生发展过程中起着促癌基因作用,为卵巢癌的深入研究提供了一条新思路。

Expression and clinical significance of Notchl in ovarian cancer

Objective：To investigate Notch l expression in ovarian epithelial tumor and the relationship between expression of Notch1 and related clinical pathology of ovarian cancer. Methods： Immunohistochemistry, real-time polymerase chain reaction and Western blot were used to detect the expression of Notchl in 109 ovarian cancer tissues,65 patient-matched oppo-site side normal ovarian tissues and 48 normal tissues. Results ： （ 1 ） The positive rate of Notehl expression in ovarian carcinoma 95.4% （ 104/109 ） ] was significantly higher than that of matched control group 7.7 % （5/65） ] and normal control group 6.3 % （ 3/48 ） ] （ P 〈 0.01 ）. The two control groups had no significant difference. （2）The expression of Notch1 in ovarian cancers was correlated with differentiation status and FIGO stage （ P〈0.05 ）, but not with age, family history, tumor location, ascites, adjacent tissue infiltration, lymphatic metastasis and sub-types. Ovarian cancer with higher clinical patologic stage and lower differentiation degree was associated with higher expression of Notch1. Conclusions：Notchl might play an oncogenic role in the development of ovarian cancer and provide a new idea for further study of ovarian cancer.
