氯化甲基汞及三氧化二砷对K562细胞凋亡的影响

摘    要:目的研究氯化甲基汞(MMC)及三氧化二砷(ATO)对K562细胞凋亡的影响。方法K562细胞经1.25、2.5、5、10和20μmol/LMMC或ATO作用24h,采用FITC-“Annexin”V/PI双染法进行细胞凋亡率的检测。结果K562细胞接触不同剂量MMC或ATO 24h,随着剂量增加凋亡率不断增加,5.0μmol/L的细胞凋亡率分别为(31.54±6.35)%和(25.37±7.68)%,与空白对照组(10.36±3.23)%比较差异有统计学意义(P〈0.05)。结论MMC及ATO均能促进K562细胞凋亡,且呈剂量依赖性。

Abstract:Objective To study the effect of methylmercury chloride(MMC) or trioxide arsenic (ATO) on the apoptosis of K562 cells. Methods MMC or ATO treated K562 ceils in 1.25,2. 5,5,10 and 20 μmol/L for 24 h, cell apoptosis rate was detected by the way of FITC - "Annexin" V/PI. Results With the dose of MMC increasing, the apoptosis rates of K562 cells increased. The apoptosis rate of cells treated with 5.0 μmol/L MMC or ATO for 24 h were(31.54 ±6. 35)% and(25.37 ± 7. 68 )%, compared with control group ( 10. 36 ± 3.23 )%, there was significant difference ( P 〈 0. 05 ) . Conclusion MMC and ATO can induce apoptosis of K562 cells in dose - dependent manner.
