玉米低聚糖阿魏酸酯体外抗氧化作用及机制研究

摘    要:目的观察从玉米麸皮中提取的低聚糖阿魏酸酯(corn feruloyl oligosaccharides,CFOs)体外对H2O2诱导损伤的PC12细胞的保护作用,探讨其抗氧化作用及机制。方法采用H2O2建立PC12细胞损伤模型,应用MTT法检测各组PC12细胞的增殖率;倒置显微镜下观察细胞形态的变化;流式细胞术测定细胞凋亡率;试剂盒检测PC12细胞内及培养液中LDH、SOD、MDA活力及含量。结果除25μmol/L CFOs组外,其余CFOs各组的细胞活力(A值)与H2O2组相比,差别均有统计学意义(P<0.05),且随着CFOs浓度的加大,细胞活力有逐渐增强的趋势。在800μmol/L时,细胞存活率达到最高值。CFOs的抗氧化作用强度与其浓度存在量效及时效双重依赖性关系。与H2O2组相比,CFOs低、中、高浓度组细胞形态均有所好转,随浓度升高,贴壁细胞数量明显增加,细胞突触逐渐增长并趋于正常。细胞凋亡率均低于H2O2组,且具有浓度依赖性,细胞内外MDA含量明显减少;细胞液中LDH活力明显减弱;细胞内SOD活力显著增强(P<0.01)。结论 CFOs对经H2O2诱导的PC12细胞具有保护作用,该作用的机制与提高PC12细胞的抗氧化能力相关。

PILOT STUDY ON THE ANTIOXIDATION ACTIVITY IN VITRO AND MECHANISM OF FERULOY OLIGOSACCHARIDES EXTRACTED FROM MAIZE

Abstract:Objective To observe the antioxidant activity in vitro of feruloyl oligosaccharides(CFOs) extracted from maize on injury of pheochromocytoma cells(PC12) induced by hydrogen peroxide(H2O2),and the possible mechanisms.Method The cell model of PC12 injured by H2O2 was established,and cell viability was tested by MTT assay and morphological changes were observed by reversal phase microscope.Cell cycle distribution and apoptosis were determined by flow-cytometry.Lactate dehydrogenase(LDH),malondialdelyde(MDA) and superoxide dismutase(SOD) in the culture medium and MDA,SOD in cells were measured by colorimetry.Results The difference of the cell viability between CFOs groups except 25 μmol/L and H2O2 group was significant(P<0.05).The cell viability came to the highest point in the group with CFOs 800 μmol/L.The antioxidant activity in vitro of CFOs was not only in dose but also in time dependent manner.Compared with H2O2 group,the cells shape of CFOs groups returned to normal.With the increase of CFOs concentration,the amount of cells adhering to the bottom of wells was increased,and the synapse elongated too.The apoptosis rate of CFOs groups were lower than that of H2O2 group(P < 0.01),and the effect is in dose dependent manner.CFOs at different concentration could decrease the release of LDH and MDA,and enhance SOD ctivity significantly(P<0.01).Conclusion CFOs has protective effect on PC12 injured by H2O2,and the mechanisms s may be related to enhancing the antioxidant abilities of PC12.
