顺铂诱导肝癌细胞凋亡过程中的Survivin表达变化及意义

目的：研究顺铂体外对肝癌细胞株HepG2 Survivin基因表达的影响。方法：培养人肝癌细胞株HepG2,加入2μg/ml的顺铂,对照组和加药后24h、48h分别以PT-PCR和免疫印迹技术检测Survivin mRNA和蛋白的表达,流式细胞仪检测细胞凋亡率变化。结果：顺铂体外对肝癌HepG2细胞作用后,对照组和加药后24h和48h的细胞凋亡率分别是（1.74±0.36）%,（9.38±1.10）%,（16.87±2.26）%;其Survivin mRNA则分别比对照组提高了1.3倍和2.8倍;蛋白分别提高了1.2倍和3.5倍。结论：顺铂可促使肝癌细胞内Survivin的表达上调,抵抗其诱导的细胞凋亡。

The alteration and significance of Survivin expression in hepatocellular carcinoma cell apoptosis induced by Cisplatin

Objective：To investigate the alteration of Survivin gene expression in hepatoma carcinoma cell line HepG2 after Cisplatin chemotherapy in virto.Methods：Cultivate the human hepatoma carcinoma cell line HepG2,and give 2μg/ml of Cisplatin to these cells for 24h or 48h,then Survivin mRNA and protein expression level was evaluated by PT-PCR and Western blot in HepG2 cells and chemotherapy group respectively.Meanwhile,HepG2 cell apoptosis rate was detected by flow cytometry.Results：The apoptosis rate of HepG2 cell and chemotherapy group with Cisplatin for 24h、48h were（1.74±0.36）%,（9.38±1.10）%,（16.87±2.26）% respectively;compared with control group,Survivin mRNA level was increased 1.3 and 2.8 time,Survivin protein level was increased 1.2 and 3.5 time with Cisplatin for 24h and 48h respectively.Conclusion：Chemotherapy with Cisplatin can enhance the Survivin expression in hepatoma carcinoma cell,and then resist the apoptosis induced by chemotherapy agent.
