摘    要:目的:观察rHuEpo对原代培养的子宫内膜异位症异位子宫内膜细胞生物学行为的影响.方法:培养原代子宫内膜异位症异位子宫内膜细胞及正常子宫内膜细胞,分为4组:异位内膜细胞缺氧组、正常内膜细胞缺氧组、异位内膜细胞rHuEpo干预组(浓度50mIU/ml)、异位内膜细胞对照组.观察各组细胞生长活性,Epo - ELISA检测各组细胞上清液中Epo浓度,流式细胞仪检测细胞凋亡比率,超微结构观察.结果:rHuEpo干预组细胞生长趋势好于单纯缺氧组细胞,凋亡比率较低(5.3%).缺氧处理后,异位子宫内膜细胞产生较高浓度的Epo( 14.2±1.7) mIU/rnl,明显高于正常子宫内膜细胞(9.1±1.5mIU/ml,q=13.48,P<0.01).结论:rHuEpo可以抑制缺氧诱导的异位子宫内膜细胞的凋亡;缺氧可以诱导子宫内膜细胞Epo表达增高.

Abstract:Objective:To study the effect of rHuEpo on ectopic endometrium cell.Methods: Ectopic endometrium cell and eutopic endometrium cell were primarily cultured,then divided into 4 groups: anoxic group of ectopic endometrium cell,anoxic group of eutopic endometrium cell,rHuEpo-interfering group of ectopic endometrium,control group of ectopic endometrium cell.The growth curve was made,and the concentration of Epo in cultural medium was detected by ELISA.Moreover the apoptosis rate and ultrastructure of every group were observed.Results: Ectopic cell affected by rHuEpo grew better than anoxic cell group,and its apoptosis rate was lower(5.3%).Ectopic cell could produce more Epo(14.2±1.7mIU/ml) after anoxic treatment,which was higher tnan eutopic cell(9.1±1.5mIU/ml,q=13.48,P<0.01).Conclusion: rHuEpo could inhibit the apoptosis of ectopic cell induced by hypoxia.Hypoxia could induce endometrium cell to produce Epo.
