慢病毒载体介导的Annexin-A10对HepG2皮下移植瘤的抑制作用

目的:观察重组慢病毒对人肝癌裸鼠皮下移植瘤体生长情况的影响及其对移植瘤细胞中膜联蛋白A10（Annexin-A10，ANXA10）、基质金属蛋白酶-9（matrix metalloproteinase-9，MMP-9）表达的影响。方法:制备HepG2细胞悬液注入裸鼠右侧腋下。待瘤体直径长至1~2cm时，取出瘤体，分别接种至24只雌性裸鼠右侧腋窝皮下。每3天测量瘤体大小，计算瘤体体积。待瘤体直径长至0.3~0.5cm时，将裸鼠随机分为3组，分别为实验组、阴性对照组及空白对照组。3周后处死裸鼠，并取出裸鼠皮下瘤体。采用免疫组化法检测ANXA10、MMP-9的表达。结果:实验组裸鼠皮下移植瘤生长明显受抑制，体积和重量均明显小于阴性对照组和空白对照组（体积:F=19.13，P<0.01;重量:F=46.91,P<0.01）。免疫组化结果显示:实验组裸鼠瘤体较阴性对照组及空白对照组的ANXA10蛋白表达明显升高；同时，MMP-9蛋白表达明显下降，差异有统计学意义（P<0.05）。结论:重组慢病毒抑制人肝癌裸鼠皮下移植瘤体的生长，ANXA10基因过表达的慢病毒抑制MMP-9蛋白的表达。

Intratumoral injection of recombinant lentivirus mediated Annexin -A10 inhibits the growth of HepG2 xenografts in nude mice

Objective:Initially illuminated the effects of ANXA10 gene on xenografts in nude mice of cell prolifer-ation,apoptosis and metachoresis.Methods:Human hepatocellular carcinoma HepG2 cell suspension was injected into right subcutaneously in nude mice,when the tumor diameter up to 1 ~2cm,were inoculated into the right subcutane-ously of the 24 female nude mice and measuring tumor size in every 3d to calculate the tumor volume.Mice were di-vided into experimental group,negative control group and blank control group randomly.Nude mice were sacrificed and tumor samples removed later.ANXA10 expression in tumor samples were dectected by immunohistochemistry.Re-sults:The growth rate of subcutaneous xenograft significantly decreased in experimental group,the tumor's size and weight of experimental group were significantly lesser than those of negative control group and blank control group(For volume,F =19.13,P <0.01.For weight,F =46.91,P <0.01).The expression level of ANXA10 protein was higher than the other two groups(P <0.05).In negative control group and blank control group,the expression level of MMP-9 protein was higher than the experimental groups(P <0.05 ).Conclusion:The overexpression of ANXA10 in HepG2 xenografts maybe inhibited the expression of MMP -9,which were the cytokines related to liver cancer inva-sion and metastasis.
