摘    要:目的:检测结肠癌细胞系SW480及正常黏膜细胞中microRNA-483(miR-483)的差异表达,探索miR-483的表达对SW480细胞生长和耐药性的影响.方法:采用实时荧光定量PCR技术比较结肠癌细胞及正常黏膜细胞中miR-483的表达差异.通过MTT法及裸鼠成瘤实验探索miR-483对SW480细胞生长的调控作用.通过流式细胞仪检测阿霉素ADR诱导的SW480细胞凋亡.结果:miR-483在结肠癌细胞中的表达升高.下调miR-483不仅抑制SW480细胞的体内外增殖能力,还能增强SW480细胞的药物敏感性.结论:miR-483在结肠癌细胞中高表达,且在结肠癌细胞增殖和耐药中发挥调控作用.

Abstract:Objective:To investigate the expression levels of microRNA-483 (miR-483) in SW480 and normal mucosa cells,and to investigate the role of miR-483 in the proliferation and drug resistance of SW480 cells.Methods:Quantitative real-time PCR (qRT-PCR) was performed to analyze the expression levels of miR-483 in SW480 and normal mucosa cells,and the correlation of miR-483 expression with the proliferation of SW480 cells was analyzed by in vitro and in vivo assay.Cell apoptosis was analyzed by flow cytometry.Results:The expression level of miR-483 in colon cancer cells was significantly higher than in normal mucosa cells.Down-regulation of miR-483 could not only suppress the proliferation of SW480 cells,but also decrease the tumor growth in mice.The flow cytometry analysis showed that the rate of ADR induced apoptosis in SW480 cells was significantly increased by down-regulating miR-483 expression.Conclusion:The expression of miR-483 was significantly upregulated in colon cancer cells.miR-483 may play an important role in the proliferation and drug resistance of colon cancer cells.
