摘    要:目的:探讨miR-21-5p通过靶向FGF18促进人鼻咽癌CNE1细胞增殖的具体机制。方法:Real-time quantitative PCR检测miR-21-5p、FGF18在人鼻咽癌组织及各鼻咽癌细胞系中的表达情况。对照组转染mimics/inhibitor NC,实验组转染miR-21-5p mimics/inhibitor/siFGF18。利用MTT、细胞克隆形成实验观察miR-21-5p及FGF18对CNE1细胞增殖活力的影响;Luciferase实验验证miR-21-5p和FGF18 3'-UTR的结合;Western blot检测FGF18、PI3K及Akt等的表达情况。结果:RT-PCR检测:人鼻咽癌组织及其细胞系中miR-21-5p及FGF18表达均显著增加(P<0.05)。MTT及细胞克隆形成实验:CNE1细胞瞬转miR-21-5p inhibitor后活细胞数量明显减少,增殖能力显著下降(P<0.05);而在miR-21-5p inhibitor+siFGF18组中,这种增殖抑制作用显著增强(P<0.05)。Luciferase实验:miR-21-5p与FGF18 3'-UTR直接结合。Western blot检测:miR-21-5p inhibitor组FGF18、PI3K及Akt表达显著减少(P<0.05)。结论:miR-21-5p靶向结合FGF18基因,激活PI3K/Akt信号通路,从而促进人鼻咽癌组织及CNE1细胞的增殖。

Abstract:Objective:To explore the specific mechanism of miR-21-5p promoting the proliferation of human nasopharyngeal carcinoma CNE1 cells through targeting FGF18.Methods:Real-time quantitative PCR was used to detect the expression of miR-21-5p and fibroblast growth factor 18 in human nasopharyngeal carcinoma tissues and cell lines.The control group was transfected with mimics/inhibitor NC,and the experimental group was transfected with miR-21-5p mimics/inhibitor/siFGF18.The effects of miR-21-5p and FGF18 on the proliferation of CNE1 cells were observed by MTT and clonogenic assay.Luciferase experiments verify the combination of miR-21-5p and FGF18 3'-UTR.Western blot was used to detect the expression of FGF18,PI3K and Akt.Result:RT-PCR assay showed that the expression of miR-21-5p and FGF18 in human nasopharyngeal carcinoma tissues and cell lines was increased significantly (P<0.05).MTT assay and cell clone formation assay showed that the number of viable cells and proliferation ability of CNE1 cells was significantly decreased by transient microarray-21-5p inhibitor (P<0.05).In miR-21-5p inhibitor+siFGF18 group,the proliferation inhibition was significantly enhanced (P<0.05).Luciferase experiment showed miR-21-5p was directly combined with FGF18 3'-UTR.Western blot detection showed the expression of FGF18,PI3K and Akt decreased significantly in miR-21-5p inhibitor group (P<0.05).Conclusion:miR-21-5p targets FGF18 gene and activates PI3K/Akt signaling pathway.It can promote the proliferation of human nasopharyngeal carcinoma and CNE1 cells.
