肿瘤组织和外周血细胞肿瘤转移抑制基因表达的对比研究

摘    要:目的:探讨恶性肿瘤患者肿瘤组织和外周血细胞中肿瘤转移抑制基因表达的相关性。方法:采用流式细胞术对恶性肿瘤患者肿瘤组织和外周血细胞的nm23、DCC、p16和p53V细胞检出率进行检测分析。结果:恶性肿瘤患者的肿瘤组织和外周血细胞nm23、DCC和p16细胞的检出率均显著低于正常对照组P<0·01和P<0·05。肿瘤转移者nm23、DCC和p16的表达率均显著低于未转移者P<0·01或P<0·05。p53V细胞检出率的变化规律与nm23、DCC、p16相反,且差异有统计学意义,P<0·01。肿瘤患者癌组织和外周血细胞之间的nm23和p53V基因编码蛋白表达率均呈良好的正相关关系,r1=0·3652,P1=0·0073;r2=0·3321,P2=0·0091,而p16和DCC的表达率无显著相关性。结论:肿瘤转移抑制基因表达与肿瘤转移呈相关性,检测肿瘤患者外周血细胞肿瘤转移基因nm23、p53的表达有助于肿瘤预后的判断。

Abstract:OBJECTIVE:To study the relationship of the expression level of tumor metastasis-suppressor genes between tumor tissues and peripheral blood cells from patients with malignant carcinomas. METHODS: The expression rates of nm23 , DCC , and p16 and p53V cells in tumor tissues and peripheral blood cells from patients with malignant carcinomas were measured and analyzed by flow cytometry. RESULTS:The expression rates of nm23 , DCC and p16 cells in tumor tissues and peripheral blood cells from patients with malignant carcinoma were lower than those of normal control groups,P<0.01 or P<0.05. The expression rates of nm23 , DCC and p16 cells in tumor tissue and peripheral blood cells from patients with metastasis were lower than those of patients without metastasis, P<0.01 or P<0.05. The regularity of expression rates of p53 were contrary comparable with nm23, DCC, p16 and these were significantly different, P<0.01. There were significantly positive correlations (r_1=0.365 2,P_1=0.007 3; r_2= 0.332 1,P_2=0.009 1) of nm23 and p53V expression rate between tumor tissues and peripheral blood cells,while p16 and DCC not. CONCLUSIONS: Tumor metastasis-suppressor genes are closely related to tumor metastasis. Determination of expression level of nm23 or p53V in peripheral blood cells may be regarded as a substitute of those in tumor tissues for the prognosis judgement of patients with malignant tumor.
