钠尿肽受体A在食管鳞状细胞癌中的表达及对食管癌细胞迁移和侵袭能力的影响

摘    要:目的探讨钠尿肽受体A(NPRA)在食管鳞状细胞癌中的表达及其对食管癌细胞迁移和侵袭的影响。方法采用免疫组织化学染色法检测119例食管鳞状细胞癌组织和91例癌旁组织中NPRA的表达情况,并分析食管鳞状细胞癌组织中NPRA表达情况与患者临床特征及预后的关系。采用蛋白质印迹法(Western blot)检测食管癌细胞株Eca109、TE-1以及正常食管上皮细胞株Het-1A中NPRA的表达情况,采用短发夹RNA(shRNA)沉默食管癌细胞株Eca109细胞中NPRA基因表达,应用Transwell小室实验分析其对Eca109细胞迁移和侵袭的影响。结果 NPRA在食管鳞状细胞癌组织中的高表达率为69.7%(83/119),明显高于癌旁组织的27.5%(25/91),差异有统计学意义(P﹤0.01)。分化程度为G2~G3级、TNM分期为Ⅱ~Ⅲ期的食管鳞状细胞癌患者食管鳞状细胞癌组织中NPRA的高表达率分别明显高于分化程度为G1级、TNM分期为Ⅰ期的患者,差异均有统计学意义(P﹤0.01)。NPRA低表达患者的5年生存情况优于NPRA高表达患者,差异有统计学意义(P﹤0.05)。食管癌细胞株Eca109、TE-1中NPRA的表达水平均高于正常食管上皮细胞株Het-1A,差异均有统计学意义(P﹤0.05)。shRNA沉默食管癌细胞株Eca109中NPRA基因的表达后,肿瘤细胞的迁移和侵袭能力明显降低。结论 NPRA在食管鳞状细胞癌组织中表达水平较高,且其高表达与肿瘤的TNM分期、分化程度以及预后密切相关,同时可以促进食管癌细胞的迁移与侵袭。

Abstract:Objective To investigate the expression of natriuretic peptide receptor A(NPRA)in esophageal squamous cell carcinoma and its effect on the migration and invasion of esophageal carcinoma cell.Method The expression of NPRA m 119 cases of esophageal squamous cell carcinoma tissues and 91 cases of adjacent tissues were detected by immunohistocheimcal staining,and the relationship between NPRA expression in esophageal squamous cell carcinoma tissues and the clinical characteristics and prognosis of relevant patients were analyzed.The expression of NPRA m esophageal carcinoma cell lines Ecal09,TE-1 and normal esophageal epithelial cell line Het-1 A were measured by Western blot.Short hairpin RNA(shRNA)was used to silence NPRA in esophageal carcinoma cell line Ecal09,and Transwell experiment was used to present its effect on the migration and invasion of Eca109 cells.Result The high expression rate of NPRA in esophageal squamous cell carcinoma tissues was 69.7%(83/119),which was significantly higher than 27.5%(25/91)in adjacent tissues,the difference was statistically significant(P<0.01).The high expression rate of NPRA in esophageal squamous cell carcinoma tissues of patients with differentiation degree G2-G3 and TNM stageⅡ-Ⅲwas significantly higher than that in patients with differentiation degree G1 and TNM stage I,and the differences were statistically significant(P<0.01).The 5-year overall survival of patients with low NPRA expression was better than that of patients with high NPRA expression,and the difference was statistically significant(P<0.05).The expression level of NPRA in esophageal carcinoma cell lines Eca109 and TE-1 were higher than that in normal esophageal epithelial cell line Het-1 A,and the differences were statistically significant(P<0.05).After transfecting shRNA to silent the expression of NPRA gene in esophageal carcinoma cell line Eca109,the migration and invasion ability of tumor cells were significantly reduced.Conclusion NPRA is highly expressed in esophageal sauamous cell carcinoma tissues,and its high expression is closely related to the TNM stage,degree of differentiation and prognosis,and it can promote the invasion and migration of esophageal carcinoma cell.
