| 摘 要： | 目的研究红细胞密度与天龄的相关性,不同密度红细胞所对应的平均天龄。方法采用非连续密度梯度分离方法,将全血红细胞分为6个不同密度的红细胞部分。测定不同部分红细胞丙酮酸激酶活性,通过各部分丙酮酸激酶活性与全血红细胞丙酮酸激酶活性的比值(Ratio)推测各部分红细胞平均天龄;测定各部分红细胞膜表面磷脂酰丝氨酸(PS)表达阳性率,来反映红细胞密度与衰老(天龄)的相关性。结果根据红细胞比值,各部分红细胞平均天龄由低密度至高密度依次为11.5、466、3.87、4.6、80.5、102.2 d。随着红细胞密度的增加,PS表达阳性率也逐渐增高。结论不同密度的红细胞代表不同的细胞天龄,采用密度梯度分离方法可以分离出不同天龄段的红细胞,为进一步研究红细胞天龄与功能的相关性提供了方法学基础。
