摘    要:目的探讨女性乳腺癌组织中雄激素受体(AR)的表达与启动子甲基化的关系。方法应用免疫组化和实时荧光定量PCR方法检测38例乳腺癌组织中AR表达及其启动子甲基化情况,并结合临床病理资料进行分析。结果乳腺癌组织中AR表达和年龄、淋巴结转移、组织学分级、ER及HER-2表达无显著相关性(P0.05);AR表达阴性组AR基因甲基化水平显著高于阳性组(P=0.015),患者年龄与AR基因甲基化水平呈负相关(r=-0.3486,P=0.0372);淋巴结转移、组织学分级及ER与AR基因甲基化无显著相关性(P0.05)。结论 AR表达阴性的乳腺癌患者AR基因甲基化水平较高。

Relationship between androgen receptor expression and its promoter methylation in patients with breast cancer Wang Hai,Ling Yang,Sheng Guifeng,Liu
Abstract:Objective To investigate the relationship between the expression of androgen receptor (AR)and promoter methylation in patients with breast cancer.Methods AR protein expression and promoter methylation status of 38 cases with breast cancer were detected by immunohistochemistry and real-time fluorescence quantitative PCR,and the clinical pathological data were analyzed.Results There were no significant correlation between AR expression and age, lymph node metastasis,histological grade,ER and HER-2 expression in breast cancer(P >0.05).The methylation level of AR gene in AR expression negative group was significantly higher than that in the positive group(P =0.015).The age of the patients was negatively correlated with the methylation level of AR gene(r =-0.3486,P =0.0372).There were no significant correlation between lymph node metastasis,histological grade,ER and AR gene methylation (P >0.05).Conclusion The methylation level of AR gene is significantly higher in patients with negative-AR breast cancer.
