摘    要:目的探讨不同化疗药物在体外培养的经TAM同步化MCF-7细胞存活率和凋亡的影响。方法以体外培养雌激素受体阳性细胞MCF-7为研究对象,采用MTr法和流式细胞术法检测MCF-7细胞存活率和细胞凋亡。结果体外培养的MCF-7细胞的倍增时间是33h,TAM可使MCF-7同步化于G0/G1期,5.Fu、DDP、ADM在中低剂量时可使同步化的MCF-7细胞存活率降低,与不同步化组比较差异具有显著性(P〈0.05)。采用流式细胞术方法检测DDP、ADM中低剂量组可使经TAM同步化的MCF-7细胞PCD%增加,与不同步化组相比较差异具有显著性(P〈0.05)。结论不同化疗药物可诱导经TAM同步化的细胞发生凋亡。

Abstract:Objective To study the effect of different chemotherapy medicine on the survival rate and apoptosis of synchronized MCF-7 cells in culture.Methods MTT assay and flow cytometer(FCM) assay were used to determine the survival rate and apoptosis of MCF-7 cells,which was estrogen receptor positive in culture.Results The time of MCF-7 cells to proliferate twice was 33 hours,and TAM could synchronize the MCF-7 in G0/G1 phase.When using MTT assay,it was found that 5-Fu,DDP,ADM could decrease the survival rate of MCF-7 in middle-and low-dose groups,and had a significant difference compared with the asynchronized groups(P<0.05).When using FCM assay,it was determined that DDP,ADM could increase the PCD% of MCF-7 in middle-and low-dose groups and had a significant difference compared with the asynchronized groups(P<0.05).Conclusion Different chemotherapy medicine can induce the apoptosis of the MCF-7 cells.
