猪带绦虫乳酸脱氢酶A和B的生物信息学比较分析

摘    要:目的预测及比较分析猪带绦虫乳酸脱氢酶A(Taenia solium lactate dehydrogenase A,TsLDH-A)和乳酸脱氢酶B(Taenia solium lactate dehydrogenase B,TsLDH-B),用于指导其生物学功能的研究。方法利用生物信息网站如美国国家生物技术信息中心(NCBI,http://www.ncbi.nlm.nih.gov/)和瑞士生物信息学研究所的蛋白分析专家系统(ExPASY,http://ca.expasy.org/)中有关基因和蛋白的序列和结构信息分析的各种工具,结合其它生物信息学分析软件包,从猪带绦虫成虫全长cDNA质粒文库中识别LDH-A和LDH-B的全长编码基因并对其结构与功能进行生物信息学预测分析。结果两序列都是包含完整开放阅读框的全长基因,推导出的氨基酸序列与其它物种LDH-A或LDH-B同源基因的氨基酸序列的一致性均大于50%。两者编码的蛋白在编码的氨基酸数目(331)、蛋白的理化性质、L-乳酸脱氢酶结构域、构成LDH酶催化中心的关键氨基酸、包含LDH活性位点的线性表位、无亚细胞定位等方面是一致的,但两者在翻译后的修饰位点、3个跨膜区和其他线性表位方面既相似也有区别。结论应用生物信息方法从猪带绦虫成虫cDNA文库中筛选出了TsLDH-A和TsLDH-B的cDNA全长序列,并预测和比较了两者结构与功能方面的信息,为进一步研究所编码蛋白的功能奠定了基础。

Abstract:Objective To predict and compare the structure and characteristics of the the genes encoding LDH-A and LDH-B protein from Taenia solium by bioinformatics in order to guide the experimental research for their biological function and application.Methods Bioinformatics analyzing tools in the bioinformatics webs site such as NCBI(http://www.ncbi.nlm.nih.gov /),ExPaSy(http://www.expasy.org /),IEDB Analysis Recource(http://tools.immuneepitope.org /main /jsp /menu.jsp) and combining bioinformatics software package,were used to identify two full-length genes encoding LDH-A and LDH-B protein from the T.solium full-length cDNA plasmid library.Results were used to predict and analyze the characteristics of the two deduced proteins.Results The two cDNA sequences are both full-length genes containing ORF of 993 base pairs encoding a putative protein of 331 amino acids.BLASTx showed that these two genes had more than 50% identity to LDH-A or LDH-B,respectively.The two deduced proteins are similar in terms of the number of encoded amino acids,stable physical-chemical characteristics,having a conserved LDH domain and an active site of LDH,the key residues in the catalytic site of enzyme,the Bcell epitopes containing LDH active site,and the lack of sequence for subcellular localization.And they are different in the modification sites after translation,three transmembrane regions and other epitopes.Conclusion Two genes encoding TsLDH-A and TsLDH-B proteins were screened out from the cDNA library of adult T.solium by bioinformatical methods.Their structure and function were predicted,analyzed and compared.
