免疫层析法快速检测动物源性食品中五氯酚酸钠含量

摘    要:目的建立胶体金免疫层析法快速检测动物源性食品中五氯酚酸钠的检测方法,对不同基质采用与其相适应的提取方法及净化方式,实现食品中五氯酚酸钠的现场快速筛查。方法样品经80%乙腈水溶液(含8%三乙胺)提取,猪肉、牛肉等肌肉不经净化、动物内脏及鸡蛋采用净化包(300mg无水硫酸钠+100mg C;+100 mgN-丙基乙二胺)净化、鱼肉采用乙腈饱和的正己烷净化,净化后的提取液经浓缩、复溶、孵育,最后用五氯酚酸钠胶体金检测卡进行检测。结果该方法检出限为1.0μg/kg,2,4-二氯酚、2,4,6-三氯酚和苯酚对五氯酚酸钠的检测结果没有明显干扰。通过对样品进行加标检测,其结果与现行国家标准检测结果一致,并且20次重复结果一致,对46份样品进行检测,结果均为阴性,且免疫层析法结果与现行国家标准方法检测结果一致。结论本方法具有简便、快速、低成本等特点,适合大批量样品的现场筛查,提高监管效率。

Abstract:Objective To establish a method for the rapid detection of sodium pentachlorophenolate in animal-derived foods by colloidal gold immunochromatography,by changing appropriate extraction methods and purification methods for different substrates to realize on-site rapid screening of sodium pentachlorophenolate in foods.Methods The samples were extracted with 80%acetonitrile aqueous solution(including 8%triethylamine),and the pork,beef and other muscles were not purified,animal viscera and eggs were purified with purification bag(300 mg anhydrous sodium sulfate+100 mg C;+100 mg primary secondary amine),fish meat was purified with acetonitrile saturated n-hexane.After purification,the extracted solution was concentrated,redissolved and incubated,and finally detected with sodium pentachlorophenolate colloidal gold detection card.Results The limit of detection of this method was 1.0μg/kg,2,4-dichlorophenol,2,4,6-trichlorophenol and sodium p-pentachlorophenolate had no significant interference with the test results.Through testing the actual samples,the results were consistent with the current national standard,and the results of 20 repetitions were consistent.On the other way,46 samples were tested,the results of immunochromatography were consistent with the test results of the current national standard method.Conclusion This method is simple,rapid and low-cost.It is suitable for on-site screening of large quantities of samples and improving the supervision efficiency.
