紫阳富硒茶中硒蛋白的提取

摘    要:为了优化碱法提取硒蛋白的工艺条件,在单因素试验的基础上,通过Box-Benhnken中心组合试验设计及响应面分析方法,考察了碱液浓度、提取温度以及料液比三因素对蛋白质提取率的影响,得出了硒蛋白提取效果的回归模型。结果表明,最佳工艺参数为碱液浓度0.09 mol/L,提取温度66.81℃,料液比1∶54.98(g:mL),每次提取3 h,提取2次,在此基础上硒蛋白提取率为56.52%,硒含量为4.89μg/g,占茶叶总硒含量的41.0%。

Abstract:In order to optimize extraction condition of Selenium-protein,according to the analysis method of center combination test and response surface methodology,the effects of NaOH concentration,extraction temperature and ratio of solid/liquid on Selenium-protein extraction were studied on the basis of the results of single factor tests,the regression model of the extraction effect of Selenium-protein was established.The optimum technology conditions were confirmed as follows: concentration of NaOH slurry 0.09 mol/L,extraction temperature 66.81 ℃, ratio of solid/liquid 1∶54.98(g:mL),extracting 3 h each time for 2 times.Under this condition,the yield of Selenium-protein was 56.52% and the content of selenium was 4.89 μg/g,accounting for 41.0% total selenium in tea.
