基于UPLC-Q-Orbitrap-HRMS分析油茶果不同部位化学成分

摘    要:基于超高效液相色谱-四级杆/静电场轨道阱高分辨质谱(UPLC-Q-Orbitrap-HRMS)技术,对“黔玉1号”油茶果不同部位化学成分进行分析鉴定,以促进油茶资源的开发和利用。采用70%乙醇对样品进行超声提取,通过UPLC-Q-Orbitrap-HRMS结合Compound Discoverer 3.0软件对一级质谱碎片进行分析,利用中药成分高分辨质谱数据库(OTCML)和文献对二级质谱碎片进行鉴定。采用聚类分析和主成分分析比较进行不同部位化学成分差异。共鉴定出51种化合物,包括黄酮类26种,萜类6种,有机酸类7种,苯丙素类3种,木脂素类2种、氨基酸4种、核苷1种、脂肪酸1种、其他类1种。通过主成分分析及聚类分析发现4个部位化学成分具有明显差异,筛选出了19种差异成分,主要为黄酮类化合物。外果皮中含有大量黄酮类化合物,根皮素是主要差异成分;萜类和苯丙素类化合物主要分布在内种皮中,原花青素B1和雷公藤内酯甲是主要差异成分;果壳和种仁含有氨基酸、木脂素类等成分,柠檬酸和山茶苷B是主要差异成分。

Analysis of Chemical Components in Diferent Parts of Camellia Oleifera Abel. Fruit Based on UPLC-Q-Orbitrap-HRMS
Abstract:Abstract: To qualitatively analyze chemical constituents from different parts of Camellia Oleifera Abel. fruit "Qianyu No. 1" by ultra-high performance liquid chromatography coupled with Q-exanctive quadrupole/electrostatic orbitrap high-resolution mass spectrometry (UPLC-Q-Orbitrap-HRMS) and to promote the development and utilization of Camellia Oleifera Abel. resources. The samples were extracted by ultrasonic with 70% ethanol. UPLC-Q-Orbitrap-HRMS techniques combined with Compound Discoverer 3.0 software were used to analysis of primary fragments. Database of orbitrap traditional chinese medicine library ( OTCML) and literature information were used to identify the secondary fragments. Analysis methods such as cluster analysis, rinciple component analysis were performed to evaluate the differences of chemical components in different parts. A total of 51 compounds, including 26 flavonoids, 6 terpenes, 7 organic acids, 3 phenylpropanoids, 2 lignans, 4 amino acids, 1 nucleoside, 1 fatty acid, and 1 others compounds were identified. Through cluster analysis and principal component analysis, it was found that chemical compositions of four parts were significantly different, and 19 significant different components were screened out, mainly flavonoids. The exocarp contains a lot of flavonoids, and phloretin is the main differential component; terpenoids and phenylpropanoid compounds are mainly distributed in the endocarp, and procyanidin B1 and wilforlide A are the main differential components; the husk and seed kernel contain Amino acids, lignans and other components, citric acid and camelliaside B are the main difference components. By exploring the differences in the compositions from different parts of Camellia Oleifera Abel. fruit "Qianyu No. 1", we provide theoretical basis for the development and utilization of Camellia Oleifera Abel. resources in Guizhou.
