摘    要:建立了柱前衍生反相高效液相色谱-紫外检测富硒脱水菜心中硒代胱氨酸(SeCys)、硒甲基半胱氨酸(SeMeCys)和硒代蛋氨酸(SeMet)的方法,优化了样品预处理方法,并评价其定量分析的线性、准确性、重复性和检出限,结果显示:以0.1mol/LHCl为提取溶剂,超声处理8h,提取效果最佳;3种硒代氨基酸的平均加标回收率为95.7%~112.9%;在各硒代氨基酸的线性范围内,线性关系良好(R>0.999);保留时间和峰面积重复性的相对标准偏差(RSD)均分别低于2.0%和5.0%;酸性条件下的检出限为0.004~0.009mg/L。

Abstract:A reversed phase high performance liquid chromatography/variable wavelength detector with precolumn derivatization was developed for the simultaneous determination of selenocystine(SeCys),selenomethylcysteine(SeMeCys) and selenomethionine(SeMet).This method was validated for linearity,accuracy,repeatability and limits of detection(LOD).The results showed that average recoveries of seleno-amino acids standards ranged from 95.7%to 112.9%,linearity was good(R2> 0.999);the relative standard deviations of repeatability for retention time and peak area were less than 2.0% and 5.0%,respectively;LOD was 0.004~0.009 mg/L in acid condition.The method is simple,accurate,and sensitive and reliable for quality control of enriched-selenium dehydrated Brassica Chinensis.
