摘    要:以一株产β-环糊精葡萄糖基转移酶(β-CGTase)的嗜热菌株HY15为研究对象,采用单因素试验研究碳源、氮源、初始pH值、培养温度、MgSO4·7H2O等对酶活力影响。并设计正交试验优化其发酵条件。结果表明最优的发酵条件为:碳源为玉米淀粉+糖蜜,氮源为下米浆,初始pH7.0,培养温度60℃、摇床转速220r/min,MgSO4浓度10mmol/L,在此条件下,β-CGTase酶活力可达1794.3U/mL。

Abstract:A β-cyclodextri glucanotransferase (β-CGTase) producing thermophiles HY15 was studied on its fermentation conditions in this paper. Orthogonal design based on single factor tests was applied to study the effects of carbon source, nitrogen source, initial pH value, culture temperature and MgSO4·7H2O on enzyme activity. The results showed that the optimum fermention conditions were using molasses and corn starch as carbon source, corn steep liquor as nitrogen source, initial pH value 7.0, fermentation temperature 60℃, rotation speed of shaker 220r/min. The β-CGTase activity could be up to1794.3U/ml under these conditions.
