雌马酚及大豆活性成分对人前列腺癌细胞DU145的生长影响

摘    要:目的:观察大豆异黄酮代谢产物雌马酚对人前列腺癌细胞DU145的生长影响,并在生理浓度下比较其与大豆异黄酮主要成分金雀异黄素、大豆素的体外抗增殖活性。方法:采用MTT法检测不同浓度的雌马酚、金雀异黄素及大豆素对DU145细胞增殖的影响,并以流式细胞术分析细胞周期分布情况。结果:雌马酚在1×10-6~1×10-5mol/L表现为抑制作用,金雀异黄素在1×10-7~1×10-6mol/L时表现为促增殖效应,达到5×10-5mol/L时出现抑制作用,大豆素在生理浓度下则主要表现为促增殖效应;雌马酚在1×10-6mol/L时使细胞在G0/G1期阻滞,在5×10-6~1×10-5mol/L时使S期及G2/M期细胞增多,金雀异黄素在5×10-6mol/L出现G2/M期阻滞,大豆素使S期及G2/M期细胞增多。结论:雌马酚在体外具有一定的抗前列腺癌活性;在生理浓度下,其抗癌活性可能强于金雀异黄素及大豆素。

Abstract:【Objective】To explore the effects of equol on cell growth of androgen independent human prostate cancer cell DU145 in vitro and compare its anti-proliferation activities with that of isoflavone daidzein and genistein.【Methods】DU145 cell was cultured in RPMI-1640 medium,then the growth effect and cell cycle distribution of different concentrations of equol were detected by MTT and flow cytometry assay.Moreover,a comparison of the effect among equol,daidzein and genistein were detected as well.【Results】The different levels of inhibitory action of equol and genistein were observed on DU145 cell at physiological concentration,while daidzein showed an opposite effect.Within the range of concentrations in the prostatic fluid,equol caused an accumulation of DU145 cell in G0/G1 at the concentration of 1×10^-6mol/L and in S and G2/M at the concentration of 5×10^-6~1×10^-5mol/L,daizein arrested cells in S and G2/M,while genistein arrested cells in G2/M at the concentration of 5×10^-6mol/L.【Conclusion】Equol showed potential anti-proliferative effects on DU145 cells in vitro,which was stronger than genistein and daidzein at concentrations that could be obtained naturally through dietary soy consumption also.
