外周血淋巴细胞M受体反应性的测定

从全血中分出淋巴细胞，在含钙Ｈａｎｋｓ’液或含磷酸二酯酶抑制剂的缓冲液中孵育，加Ｃａｒｂａｃｈｏｌ能使细胞内ｃＧＭＰ含量增高６６％－８０％，这种升高可被阿托品阻断，说明ｃＧＭＰ的增高是由Ｍ受体介导的特异性反应。据此建立了一种反映Ｍ受体ＣＧＭＰ系统功能状态的方法并对孵育和反应条件作了探讨，初步实验结果表明老年大鼠淋巴细胞ＣＧＭＰ基础含量显著低于青年大鼠，由ＣａｒｂａｃｈｏｌＧＩ起的ｃＧＭＰ升高反应也明显降低。

Determination of reactivity of M-cholinergic receptor/cGMP system of peripheral lymphocytes

Lymphocytes were separated from peripheral blood and incubated in Hanks'solution with calcium or buffer solution containing phosphodiesterase inhibitor IBMX. When carbachol at 1-10mmol/L was added to the incubation medium, the intracellular cGMP colltent as determined by high-sensitivity RIA was elevated by 66%- 80%. Such an elevation could be blocked by atropine and hence is a specific reaction mediated through M-receptors. A method based on this reaction was thus established to reflect the reactivity of M-receptor/cGMP system. Preliminary experiment revealed that the base level of lymphocyte cGMP as well as the elevation of cGMP induced by carbachol in aged(24-26 months old) rats were significantly lower than young rats. The methodology, especially the influence of incubation conditions was discussed.
