摘 要: | 研究穿山龙薯蓣总皂苷提取条件的确定及其分离提纯。实验中确定皂苷提取的最适酒精度、最适温度和最佳时间分别为95%、50℃和6h,总皂苷得率为2.4%。用柱层析对薯蓣皂苷单体进行分离提纯,得率为样品的19.6%。用TLC检测样品,确定得到样品为3 O {α L 鼠李糖 (1→4) α L 鼠李糖 (1→2)] β D 葡萄糖} 薯蓣皂苷元。最后用HPLC法检测其纯度为90%左右。

Separation and purification of dioscin |

Abstract: | The saponin extraction from Dioscorea nipponica was studied. The optimum extracting conditions are determined via experiment as follows: temperature, 50?℃; time, 6?h. The optimum ethanol concentration is 95% and the best the yield of whole dioscin is 2.4%. While separating dioscin from the mixture saponin by silica gel column, the yield of the purified dioscin is 19.6%. The samples are identified by TLC to be 3-O-{α-L-rha-(1→4)-α-L-rha-(1→2)]-β-D-glc}. The purity of dioscin is about 90%, detected by HPLC. |
