摘    要:研制了以SpreetaTM传感器为核心,结合液体处理系统、信号检测与控制器、计算机分析处理软件等模块的小型表面等离子共振(SPR)检测系统.运用该系统,通过在传感器表面偶联牛血清蛋白(BSA),采用直接检测法测量了不同浓度的BSA抗体溶液,得到了BSA抗体溶液的SPR传感图,建立了BSA抗体的浓度标准曲线.在传感器表面固定雌二醇(E2),采用间接检测法测量了不同浓度的E2溶液,根据E2溶液SPR响应信号的变化量,建立了E2的浓度标准曲线.实验中系统的分辨率为10-5折射率单位(RIU).对于实际的待测样品,只要测得其SPR信号的变化量,结合标准曲线就能够得出样品的浓度.

Abstract:Combining some modules including liquid treatment system, signal detection and control unit, computer analysis and processing software, a small surface plasmon resonance (SPR) detection system with SpreetaTM sensor as its core component was developed. By using this system, bovine serum albumin (BSA) antigens were immobilized on the sensitive layer of the SPR sensor and the BSA antibody solutions of different concentrations were detected with the direct assay method. A sensorgram of SPR signal about BSA antibody solutions was generated and a calibration curve was obtained. By immobilizing estradiols (E2) on the sensitive layer, E2 solutions of different concentrations were detected using the indirect assay method and a calibration curve was obtained according to the change of SPR signal. In these experiments, the system resolution was 10-5 refractive index unit (RIU). For the samples of unknown concentration, once the SPR signal change is detected, the concentration can be calculated from the calibration curve.
