无启动子转化策略用于分离组织特异性启动子的研究

摘    要:通过研究外源uidA基因在体内表达的时空特异性,利用PCR、GUS组织化学检测等技术,从以tritordeum为受体、经无启动子uidA转化策略得到的转基因供试材料中成功筛选出了外源uidA基因在花药原基特异表达的单株。结果表明,花药原基特异表达单株的内源组织特异性启动子已被外源uidA片段标定,为启动子的分离克隆提供了一种有效手段。

Abstract:Through researching the space-time expression character of inserted foreign uidA, the single strain with anther-primordium tissue-specific expression of foreign uidA was screened successfully using PCR and GUS from the transgenic materials obtained by the non-promoter transformation tactic using tritordeum as acceptor. The method provided an effective measure for separating and cloning promoter.
