摘    要:建立一种测定湖水中微囊藻毒素的高效液相色谱法(high performance liquid chromatography,HPLC)。以0.5mL/L三氟乙酸水溶液作为流动相,Agilent Eclipse XDB C18柱分离,紫外检测器(VWD)在波长238nm处检测。本方法最低检出限是0.4μg/L。微囊藻毒素标准浓度在0.4μg/mL-2μg/mL范围内,线性关系良好,相关系数r达0.998。加标浓度为0.4μg/mL-1.8μg/mL时,回收率为84.2%-92.0%,相对标准偏差在0.2%-3.2%之间。该方法灵敏度、准确度高,操作简单、安全。可用于测定水体中的微囊藻毒素。

Determination of Microcystin in Lake Water by HPLC

Abstract:In this study,we intended to develop a method for the determination of microcystin in lake water by high performance liquid chromatographic(HPLC). The trichloroacetic acid solution with 0.5 m L/L was applied as mobile phase. Microcystin was separated by Agilent Eclipas XDB C18 column and determined at 238 nm wavelength by VWD. The detection limit of the method reached 0.4μg/L. There was a good linearity within the range of 0.4μg/m L to 2μg/m L with the correlation coefficient of 0.998. The recoveries were between 84.2% and 92.0% at the spiking level of 0.4μg/m L to 1.8μg/m L,and the relative standard deviation ranged from 0.2% to 3.2%. The method built in this study is sensitive,accurate and easy to operate.
