摘    要:研制了细胞分离芯片和DNA提取芯片两种样品预处理微流控芯片,介绍该两种芯片的原理、结构和样品预处理效果。基于微过滤原理设计的闸式细胞分离芯片,可实现老鼠外周血中白细胞与红细胞的分离。DNA提取芯片是基于固相萃取原理设计,研制成Si-玻璃和Si-PDMS-玻璃两种结构的DNA提取芯片。采用深刻蚀技术在硅片上刻蚀出20μm宽方柱阵列或直径为10μm的圆柱阵列,刻蚀深度为30~150μm,微柱阵列作为提取DNA的固相载体,成功提取PCR产物中的DNA。

Abstract:Two types of sample pretreatment microfluicdic chips including cell separation chip and DNA extraction chip were developed. The principles,the structures and the sample pretreatment effects of these chips were described separately. Based on the principle of microfilter,gate-type cell sorter chip was designed,which can separate the white blood cells from the rat peri-pheral blood. Based on the solid phase extraction principle,DNA extraction microchips containing Si-glass microchip and Si-PDMS-glass microchip were achieved. 20 μm wide square posts array or 10 μm diameter pillars array were etched by using deep reaction ion etching technology as the solid phase matrix for absorption DNA. The etch depth is varied from 30~150 μm. The DNA extraction chips were used successfully for purification DNA from the PCR products.
