茚虫威对斑马鱼的急性毒性及遗传毒性

摘    要:茚虫威属噁二嗪类新型广谱高效杀虫剂,在农业生产中有着重要作用,近年来随着用量的增加,由此产生的对水生生物的影响越来越受到重视。使用斑马鱼进行茚虫威的急性毒性作用实验,并采用微核试验、彗星试验、Annexin V-FITC/PI双染色检测观察染毒后的斑马鱼是否被诱导产生遗传毒性。结果表明:在24 h、48 h、72 h、96 h,茚虫威LC50值分别为2.263mg·L-1、2.184 mg·L-1、2.184 mg·L-1、2.133 mg·L-1。茚虫威对斑马鱼急性毒性属中等毒性。96 h茚虫威浓度为1.98 mg·L-1和2.16 mg·L-1时,斑马鱼的微核率与对照组相比达差异极显著水平(P0.01)。采用彗星试验检测斑马鱼肝脏DNA损伤,结果显示斑马鱼暴露于高浓度1.98 mg·L-1和2.16 mg·L-1茚虫威后,与对照组相比,斑马鱼的DNA损伤均表现为极显著差异(P0.01)。采用Annexin V-FITC/PI双染色流式细胞仪法检测,结果表明染毒后24 h斑马鱼肝细胞出现细胞凋亡现象。实验结果进一步表明,茚虫威对斑马鱼短期染毒后诱导细胞凋亡,表现出遗传毒性。

Acute and Genetic Toxicity of Indoxacarb to Zebrafish (Brachydanio rerio)

Abstract:Indoxacarb is a new and efficient broad-spectrum oxadiazine insecticide, which plays a vital role in agricultural production. With the increase of indoxacarb dosage, more attention is paid to the effects of indoxacarb on aquatic organisms and water quality. Acute toxicity of indoxacarb to Brachydanio rerio was studied, and micro nuclear test, DNA Ladder and Annexin V and propidium iodide double staining were used to detect whether the treated B. rerio would produce apoptosis, to explore the preliminary toxicity mechanism of indoxacarb. The results showed that indoxacarb was moderately toxic to B. rerio, and 24h-LC50, 48h-LC50, 72h-LC50 and 96h-LC50 of acute toxicity were 2.263 mg?L-1, 2.184 mg?L-1, 2.184 mg?L-1 and 2.133 mg?L-1 respectively. B. rerio micronucleus rates in concentration of 1.98 and 2.16 mg?L-1 were extremely significant difference (P<0.01), showing that genetic toxicity exists in zebra fish. In the comet assay, the result showed that in the indoxacarb concentration of 1.98 and 2.16 mg?L-1, the liver DNA damage significant different were found compared to the control (P<0.01). And the result of Annexin V-FITC /PI revealed that apoptosis were happened in the liver cells after B. rerio was treated by indoxcarb for 24 h. The experimental results further proof that the short-term treated B. rerio with indoxacarb produce apoptosis, which indicate genetic toxicity of indoxacarb.
