弱光胁迫对烟草幼苗生长发育的影响和相关基因qRT-PCR分析

摘    要:为研究弱光胁迫对烟草生长发育的影响及特异性响应基因的表达,本研究测定了不同光照处理下烟草幼苗苗期天数以及农艺性状指标和光信号传导相关基因的差异表达模式,并分析了这些基因表达与烟草光信号转导间的关系。结果表明:弱光胁迫对于不同生育时期烟草的生长发育产生了极大的影响,具体表现为烟苗生育期延长,农艺指标均下降。在此过程中,钙感受器基因CAS、类钙调素蛋白基因CML19、钙依赖性蛋白激酶基因CDPK9、CDPK14以及ACRE20基因均在弱光胁迫处理后下调;然而钙/钙调素依赖性蛋白激酶基因CCaMK则出现上调;蔗糖磷酸合成酶基因SPSC和RuBisCo小亚基基因rcbS出现大幅下调;然而捕光叶绿素a/b结合蛋白基因CAB21的表达出现大幅上调。研究发现,弱光胁迫抑制了大部分响应钙素及光信号基因的表达,而仅有少数基因表现负调控。这些结果可为后续开展烟草遗传改良提供科学依据。

Abstract:To study the effect of low light intensity stress on tobacco growth and development and involved genes expression.In this study,tobacco seedling period,agronomic traits and the expression pattern of genes involved in light signal transduction in tobacco seedlings in different light treatments were determined.Then,the influence of gene expressions on light signal transduction was also analyzed.The results showed that low light stress had a great influence on the tobacco growth and development.The specific manifestations were prolonged growth period and agronomic traits declined.In this process,the calcium sensing receptor(Cas),calcium-binding protein(CML19),calcium-dependent protein kinase(CDPK9,CDPK14)and ACRE genes were down-regulated,while calcium and calcium/calmodulin-dependent protein kinase(CCaMK)gene was up-regulated.Sucrose-phosphate synthase(SPSC)and ribulose bisphosphate carboxylase small chain(rcbS)genes were both down-regulated,while chlorophyll a-b binding protein(CAB21)gene was up-regulated significantly.The results showed that low light stress inhibited the expression of most of the genes that responded to calcium and light signals,while only a few genes showed negative regulation.These results could provide scientific basic for tobacco genetic improvement in the future.
