二茂铁甲酸与鲱鱼精DNA相互作用的电化学研究

摘    要:用循环伏安、微分脉冲伏安和计时库仑等电化学方法结合紫外光谱法,研究了二茂铁甲酸(FCA)与DNA的相互作用.循环伏安实验表明FCA在玻碳电极上出现一对可逆的氧化还原峰.DNA的加入导致FCA氧化还原峰电流降低,且式电位往正向移动,表明二者可能通过沟面方式发生作用.计时库仑法表明FCA与DNA作用后,复合物扩散系数明显降低,这可能是FCA与DNA反应峰电流降低的原因.微分脉冲实验表明以FCA作为电化学探针,能在1.3×10^-5mol/L~1.0×10^-4mol/L浓度范围内对DNA进行定量检测.紫外光谱实验进一步证实了二者的相互作用,且作用模式可能够沟槽结合.

Abstract:The interaction between Ferrocenecarboxylic acid (FCA) and DNA was investigated by cyclic voltammetry, differential pulse voltammetry, chronocoulometry and UV spectrophotometry. It was showed that FCA was an electroactive compound at glassy carbon electrode with a pair of reversible redox peaks. The interaction of DNA with FCA caused the significant decrease of redox peak currents coupled with positive shift of peak potentials, indicating that FCA might bind with DNA with a groove binding mode. Chronocoulometry suggested that, after interaction with DNA, the diffnsion rate of the complex was decreased obviously, which might be the reason that caused the decrease of redox peak currents. Differential pulse voltammetry showed that the concentration of DNA could be qualified in the range of 1.3×10^-5 mol/L-1.0×10^-4 mol/L via using FCA as an electroactive probe. The interaction of FCA and DNA and their binding mode were further testified by UV absorption spectra.
