rH2AX核内斑点在不同成胶质瘤细胞中的剂量时间效应研究

摘    要:将成胶质瘤细胞MO59K和MO59J两种细胞分别给予1Gy,2Gy,4Gy和8Gy的6MV-X线照射,并分别于0.5h、1h、2h、4h、8h、12h、24h收集细胞,检测rH2AX核内斑点时间及剂量效应特点.结果发现,MO59K细胞和MO59J细胞随着照射剂量的增加rH2AX核内斑点数量增加;并且MO59K细胞接受8Gy照射后rH2AX斑点在24h内能够恢复到放疗前的水平,而MO59J细胞内rH2AX斑点不能恢复到照射前水平.提示MO59J细胞较MO59K细胞敏感,照射后rH2AX核内斑点更难恢复到未照射水平.

Abstract:We studied dose-response and time-effect relationship of rH2AX foci by immunofluorescence in different glioma cells after irradiation which then were compared for differentiation.MO59Kcell and MO59J cell along with the increase in the amount of rH2AX nuclear foci with dose increasing.MO59Kcell accepted 8Gy irradiation rH2AX foci number within 24hours can be restored to the level before the radiotherapy;The rH2AX foci in MO59Jcell within 24hours after the acceptance8Gy radiation can not be restored to the level before irradiation.The rH2AX nuclear foci is more difficult to return to the unirradiated level in MO59Jcell than in MO59Kcell after irradiation which showed that MO59Jcell is more sensitive to irradiation.
