羧基功能化超顺磁性纳米粒子吸附牛血清蛋白的特性

摘    要:以牛血清白蛋白(BSA)为目标蛋白,考察时间、BSA质量浓度、pH值、温度等条件对磁性纳米粒子吸附BSA的影响,并对吸附有BSA的磁性纳米粒子的解吸附情况进行研究.结果表明:在25~45 ℃内,温度几乎不影响吸附过程;pH值为4.0~4.5达最大吸附量,该吸附过程满足准一级动力学方程和Freundlich等温吸附模型;Na2HPO4对其有较强的脱附作用,但粒子与BSA之间不是简单的物理相互作用,而是静电相互作用,导致其脱附水平依然较低,仅为20.06%.

Abstract:Using bovine serum albumin(BSA)as the target protein, the effect of time, BSA concentration, pH, temperature on magnetic nanoparticles(MNP)of BSA was investigated, as well as the desorption of BSA from the MNP. The results showed that the temperature nearly had no effect on adsorption process when it was 25-45 ℃. It reached the max adsorption amount when pH was 4.0-4.5, and the adsorption process fitted the quasi first order kinetic equation and Freundlich isotherm adsorption model. Na2HPO4 had a strong effect on the desorption process, but the desorption rate was still low of only 20.06%. This is because it is not a simple physical interaction but the electrostatic interaction between the particle and BSA.
