普通小麦与玉米的体细胞杂交再生完整植株

摘    要:从小麦济南177制备得到两种原生质体,一种来源于具有一定分化能力的愈伤组织但其分裂能力较低;一种来源于生长迅速的悬浮细胞但不能分化,将二种原生质体混合,共同作为受体与经过紫外线(UV)照射的玉米(Zea mays L)原生质体在PEG诱导下融合,培养后获得再生克隆并进一步分化为植株,而它们单独与玉米原生质体融合均不能再生植株,通过染色体、同工酶及5SrDNA分析及生长习性分析证明再生植株均为体细胞杂种。

PRODUCTION OF PLANTS FROM SOMATIC HYBRIDIZATION BETWEEN COMMON WHEAT AND MAIZE(ZEA MAYS L)
Abstract:Two kinds of protoplast were prepared from wheat (Triticum aestivum L)cv.Jinan 177. One kind was originated from calli with rather high regeneration capacity but their protoplast possessed only low division ability.The other one was originated from fast growing cell suspensions and could not differentiate.When each kind of protoplast fused with UV-irradiated protoplasts of maize respectively,no regenerated plants could be obtained.However,when they were mixed together as receptor and fused with the same sort of maize protoplast,the fusion products grew to regenerated clones and further differentiated to plants. They were identified as hybrids by chromosome observation as well as esterase isozyme and 5S-rDNA sequence spacer analysis.
