基于RAPD的藏马鸡亲缘关系的研究

摘    要:为了测定7只笼养藏马鸡的亲缘关系,利用随机扩增多态DNA技术对7个个体进行了分析.选用53个10bp的随机引物对每只藏马鸡的基因组DNA进行扩增,获得14个有效引物,并得到226个扩增片段.根据聚类分析所得到的树状图,确定了7只藏马鸡的亲缘关系,证明RAPD可以用来鉴定亲缘关系.

Abstract:In order to determine the kinship of the offspring of the captive population that has been established in Beijing Zoo,randomly amplified polymorphic DNA(RAPD) was used to examine the parenthood of 7 Tibetan eared pheasants(Crossoptilon harmani) kept in Beijing Zoo.Fifty-three primers were selected to amplify genomic DNA of each individual.The results of amplifications show that 14 primers have clear and distinct RAPD patterns.With totally 226 amplified fragments,the cluster analysis of the 7 Tibetan eared pheasants indicate that all the 4 young birds come from the same father(No.1).This study provides a practical method to determine the relationship of birds which parents are unknown.
