摘    要:利用反相高效液相色谱法建立了同时分离和测定4'-去甲鬼臼毒素和鬼臼毒素的方法.所选用的色谱柱为Waters Spherisorb ODS2(5μm,4.6 mm i.d×250 mm),流动相组成为甲醇-水(体积比为65∶35),检测波长为207 nm.在该色谱条件下,4'-去甲鬼臼毒素质量浓度在0.222 4556μg/mL、鬼臼毒素质量浓度在0.596 8556μg/mL、鬼臼毒素质量浓度在0.596 81 492μg/mL范围内线性关系良好,相关系数分别为0.999 8和0.999 9.该方法应用于中药八角莲的叶、茎、须和根中这2个组分的含量分析.

Abstract:A reversed- phase high- performance liquid chromatography( RP- HPLC) method has been developed for the determination of 4'- demethylpodophyllotoxin( DMEP) and podophyllotoxin( PPT). The results have indicated that DMEP and PPT could be separated by a Waters Spherisorb ODS2 column( 5 μm,4. 6 mm i. d. × 250mm) at the column temperature of 40 ℃,using methanol- water( 65∶ 35,V /V) as the mobile phase with the flow rate of 1. 0 mL /min and detective wavelength of 207 nm. Under the chromatographic conditions mentioned above,the linear ranges were found as 0. 222 4—556 μg /mL for DMEP and 0. 596 8—1 492 μg /mL for PPT,with the correlation coefficients of 0. 999 8 and 0. 999 9,respectively. The method could be used for the determination of DMEP and PPT in the Dysosm aversipellis( Hance) M. Cheng.
