紫花三叉小白芨组培快繁技术研究

摘    要:[目的]确定白芨种子萌发的最适培养基,以及种子萌发后在自然光培养下丛生芽诱导分化及生根的最佳条件。[方法]选择成熟白芨果荚为外植体,采用组织培养技术对白芨种子诱导、增殖、生根等问题进行试验研究。[结果]白芨种子诱导最适宜培养基为1/2MB+马铃薯泥20 g/L+蔗糖20 g/L,诱导率为95%;增殖分化培养基为1/2MB+马铃薯泥50 g/L+香蕉泥20 g/L+6-BA0.4 mg/L+NAA 0.1 mg/L+蔗糖25 g/L+艾蒿叶干粉2 g/L,增殖系数为14.2倍;生根培养基为1/2MB+香蕉泥80 g/L+NAA0.5 mg/L+蔗糖20 g/L+艾蒿叶干粉4 g/L,生根率达100%。[结论]白芨组培快繁能培养出带根茎叶的小植株,移栽成活率可达95%,为白芨产业化发展提供技术支持。

Study on Rapid Propagation Technology of Tissue Culture of Purple Flower Trident Small Bletilla striata
Abstract:[Objective] The research aimed to determine the optimal medium for seed germination of Bletilla striata,the best conditions for inducing differentiation and rooting of multiple shoots under natural light.[Method] The mature Bletilla striata fruit pod was selected as explant,and the tissue culture technique was used to study the inducement,proliferation and rooting of Bletilla striata seed.[Result] The optimum culture medium for Bletilla striata seed induction was 1/2MB + mashed potatoes 20 g/L + sucrose 20 g/L,and the induction rate was 95%.The proliferation differentiation medium was 1/2MB + mashed potatoes 50 g/L + mashed banana 20 g/L +6-BA 0.4 mg/L + NAA 0.1 mg/L +sucrose 25 g/L + Artemisia argyi leaves dry powder 2 g/L,the multiplication coefficient was 14.2.The rooting medium was 1/2MB + mashed banana 80 g/L + NAA 0.5 mg/L + sucrose 20 g/L + Artemisia leaves dry powder 4 g/L,and the rooting rate was 100%.[Conclusion] Rapid propagation of tissue culture of Bletilla striata can cultivate plants with rhizomes and leaves,the survival rate of transplanting can reach 95%,which provides technical support for the industrialization development of Bletilla striata.
