国产玛咖芥子油苷的组分分析和含量测定

摘    要:目的:对国内不同颜色、不同地区的玛咖,及玛咖不同部位的芥子油苷的主要组分和含量进行鉴定和分析。方法:将玛咖粉末用70%甲醇提取,过DEAE-SephadexA-25阴离子交换柱后,加硫酸酯酶后于37℃的恒温箱酶解,洗脱液用LC-MS法分析其成分和含量。结果:各种不同玛咖的主要芥子油苷组分相同,均检测到苄基芥子油苷和间-甲氧基苄基芥子油苷这2种芳香族芥子油苷,其中前者占总含量约75%。芥子油苷在不同颜色玛咖中含量差异较小;在玛咖叶片、块根、须根3个不同部位含量差别很大,须根中含量高于主根,主根中高于叶片。同时,国内不同地区玛咖中芥子油苷含量也有较大差别。结论:使用LC-MS法可对国产玛咖中所含的芥子油苷组分及含量进行准确分析。

Abstract:Objective:To identify and analyse the main components and content of the glucosinolates in different parts of domestic Maca and that with different colors and from different parts.Methods:Maca powder was extracted with 70% methanol,then the extracted glucosinolates were adsorbed onto a column packed with DEAE-Sephadex A-25,followed by enzymatic hydrolysis in 37 ℃ with sulfatase.After enzymatic hydrolysis,the composition and content of the eluent can be analysed with LC-MS.Results:Main glucosinolates in various Maca have the same components,in which benzyl glucosinolates and methoxy-benzyl glucosinolates,two aromatic glucosinolates,were detected.The former occupied about 75% of the total content.Glucosinolates in Maca with different colors had little differences in content,in the three different parts(leaves,storage roots,fibrous roots) of Maca,the content of glucosinolates had significant difference,and the fibrous roots had higher content than the main roots,and the content in main roots is higher than that in leaves.In addition,the content of glucosinolates in different parts of the domestic Maca were also quite different.Conclusion:The LC-MS can be used to analyse the components and content of glucosinolates contained in Maca.
