摘    要:为了优选复方降脂口服液的提取工艺,通过网络药理学及分子对接技术分析复方降脂口服液中的功效成分,结合正交设计实验确定最佳提取工艺。结果表明,复方降脂口服液中的主要活性成分为异鼠李素、山柰酚、槲皮素等黄酮类化合物,作用于AKT1、IL6、PPARG等关键靶点,通过胰岛素抵抗、脂肪分解、AMPK等信号通路,发挥治疗单纯性肥胖的作用;以功效成分总黄酮含量及干膏得率为评价指标,正交设计试验得出最佳提取工艺条件为:焦山楂、赤小豆、决明子、茯苓、薏苡仁、枸杞子6味药加1 500 mL水浸泡60 min,煎煮45 min后下荷叶、陈皮、玫瑰花共煎25 min。可见复方降脂口服液以多成分-多靶点-多通路途径治疗单纯性肥胖,通过网络药理学及分子对接确定工艺评价指标,优选的工艺稳定,可为口服液产品开发奠定基础。

Abstract:In order to optimize the extraction process of compound lipid-lowering oral liquid, the effective components of compound lipid-lowering oral liquid were analyzed by network pharmacology and molecular docking technology, and the optimal extraction process was determined by orthogonal design experiment. The results showed that the main active ingredients in compound lipid lowering oral liquid were isohamnetin, kaempferol, quercetin and other flavonoids, which acted on key targets such as AKT1, IL6, PPARG, and played a role in the treatment of simple obesity through insulin resistance, lipolysis, AMPK and other signaling pathways. Taking the content of total flavonoids and the yield of dry paste as the evaluation indexes, orthogonal design experiment showed that the best extraction conditions were as follows: the Charred Crataegus, Vigna umbellate, cassia seed, Poria cocos, semen coicis and Chinese Wolfberry were added with 1500 mL water and steeped for 60min, decocted for 45 min and then decocted with lotus leaf, Pericarpium Citri Reticulatae and rose hips for 25 min. It can be seen that the compound lipid-lowering oral liquid could play an intervention role in the obesity by multi-component, multi-target, and multi-pathway treatment. Through network pharmacology and molecular docking, the process evaluation index is determined and the preferred process is stable which can lay the foundation for oral liquid product development.
