微泵耳蜗灌注碱性成纤维生长因子对庆大霉素致聋豚鼠的作用

摘    要:目的 :应用微量渗透泵 (MOP)向豚鼠耳蜗灌注碱性成纤维生长因子 (bFGF) ,观察其对庆大霉素中毒所致耳聋的保护作用。方法 :10只豚鼠分成庆大霉素致耳聋组及正常组 ,于 2组豚鼠右耳耳蜗植入MOP灌注bFGF ,分别于灌注前 ,灌注后 1周、2周测ABR阈值 ,扫描电镜观察耳蜗组织形态。结果 :耳聋组在灌注bFGF后 1周、2周ABR阈值较灌注前降低 (P <0 .0 5 ) ,耳蜗毛细胞形态得以恢复 ;正常组ABR阈值及形态均无明显改变。结论 :MOP持续恒速耳蜗灌注bFGF对耳蜗中毒的毛细胞有修复作用 ,对正常耳蜗功能和形态无影响

Abstract:Aim:To investigate the protective effects of bFGF perfused to cochlea by MOP on the hearing loss induced by gentamicin. Methods:Ten guinea pigs were randomly allocated into 2 groups:the deafness group and the normal control group. bFGF was perfused to the cochlea by MOP.ABR and the morphological features of cochlea before perfusion,1 and 2 week after perfusion were observed.Results:The auditory threshold of ABR decreased in the deafness group at 1 and 2 weeks after perfusion( P <0.05)and the morphological lesions of hair cell alleviated compared to those before perfusion.No sigmficant changes were fourd for normal controls.Conclusion:A continual perfusion of bFGF to cochlea by MOP can repair the hair cell injury by gentamicin and have no effects on normal cochlea.
