摘    要:为了提高微拟球藻油脂产量,采用二次正交旋转组合设计方法,选取NaNO3浓度、NaH2 PO4浓度、温度、CO2质量分数和光照强度5个因素,对微拟球藻自养产油脂的工艺参数进行优化.得到的最佳培养工艺条件为:NaNO3质量浓度0.38 g/L、NaH2PO4质量浓度0.35 g/L、温度27.1℃、CO2质量分数6.67%、光照强度5 454 lx,预测理论最大油脂产量为0.72 g/L.在最佳工艺条件下进行验证实验,实际油脂产量0.70 g/L,是优化前的3倍多.微拟球藻油脂以C16和C18脂肪酸为主.

Abstract:To increase lipid yield from Nannochloropsis oculata,five culture conditions,including NaNO3 concentration,NaH2PO4 concentration,temperature,CO2 concentration,and illumination were optimized by the method of quadratic orthogonal rotation combination experiment design.The predicted theoretical maximum lipid yield was 0.72 g/L.The optimum culture conditions of NaNO3 concentration,NaH2PO4 concentration,temperature,CO2 concentration and illumination were 0.38 g/L,0.35 g/L,27.1 ℃,6.67%,and 5 454 lx,respectively.For the verification test,a lipid yield of 0.70 g/L was achieved under the optimum conditions.It was increased by two fold more than that under the original conditions.Nannochloropsis oculata were dominated by C16 and C18 fatty acids.
