苯丁酸钠和二甲基甲酰胺对乳腺癌细胞增殖抑制影响的研究

摘    要:目的观察苯丁酸钠(PB)和二甲基甲酰胺(DMF)对乳腺癌MCF-7细胞增殖的影响。方法应用MTT比色法,以梯度浓度的PB或DMF作用于MCF-7细胞,分别于24、48、72h对细胞增殖进行检测。结果0.391~3.125mmol/LPB和0.5—8.0mmol/LDMF作用MCF-7细胞24~72h,随着PB浓度的增加或作用时间的延长,细胞生长抑制率明显增加。随着DMF浓度的增加或作用时间的延长,细胞生长抑制率也较明显增加。结论PB和DMF可有效抑制乳腺癌MCF-7细胞的增殖。

Abstract:Objective To investigate the effect of sodium phenylbutyrate and dimethylformamide on cell proliferation in breast carcinoma MCF-7 cell line. Methods MCF-7 cells grew in the presence or absence of PB and DMF for 24 h,48 h and 72 h, respectively. The cellular proliferation inhibitory ratio was evaluated by MTY assay. Results MCF-7 cells were treated by O. 391-3. 125 mmoL/L PB and 0. 5-8.0 mmol/L DMF respec- tively for 24-72 h. With the increase of concentration of PB or the prolongation of the treating time, the prolifera- tion inhibitory ratio of the tumor cells was increased notably. With the increase of concentration of DMF or the prolongation of the treating time, the proliferation inhibitory ratio of the tumor cells was also increased notably. Conclusion PB and DMF could effectively inhibit cell proliferation.
