生脉饮治疗慢性心衰大鼠不同提取工艺的比较

摘    要:目的:比较不同提取工艺的生脉饮对慢性心衰大鼠的治疗效果。方法:雄性Wistar大鼠按1.5mg.kg-1体重剂量腹腔注射阿霉素,每周2次,共7周进行造模;正常组注射相同体积的生理盐水。将造模的Wistar大鼠随机分成四组:模型组、阳性药物组、渗漉药物组、水煎药物组。连续灌胃给药4周,观察不同提取工艺制成的生脉饮对大鼠行为体征、血流动力学指标以及做心脏病理切片观察其组织学变化;检测血清中丙二醛(malondialdehyde,MDA)、乳酸脱氢酶(lactate dehydrogenase,LDH)、肌酸激酶(creatine kinase,CK)、脑钠肽(brain natriuretic peptide,BNP)、超氧化物歧化酶(superoxide dismutase,SOD)的含量。结果:与渗漉药物组相比,水煎药物组大鼠精神萎靡不振,活动量、进食量减少。水煎药物组大鼠心肌组织受损程度比渗漉药物组大。两种提取物均能降低慢性心衰大鼠血清中MDA,LDH,CK活性,升高SOD水平;渗漉提取物能极显著地降低慢性心衰大鼠血清BNP水平,水煎液则不能。渗漉药物组和水煎药物组大鼠收缩压(systolic pres-sure,SP)、舒张压(diastolic pressure,DP)、动脉血压(arterial pressure,AP)、左心室压力最大下降速率(-dp/dt-max)较模型组极显著上升。结论:治疗慢性心衰大鼠,渗漉提取工艺的效果比水煎工艺好。

Abstract:Objective:To compare the effects of Shengmaiyin prepared by different extraction methods on rats with chronic heart failure.Methods:Cardiac dysfunction was induced by intraperitoneal(IP) injection of adriamycin(ADR) into male Wistar rats.Rats with chronic heart failure were randomly divided into four groups:model group,positive medicine group,percolation extraction group,and water extraction group.The drugs were given by intragastric lavage continuously for 4 weeks.The hemodynamics indexes were measured using multipurpose polygraph and the rats were sacrificed.The hearts were processed for histological examination.The levels of malondialdehyde(MDA),lactate dehydrogenase(LDH),creatine kinase(CK),superoxide dismutase(SOD),and brain natriuretic peptide(BNP) in serum were determined.Results:Compared with the rats in percolation extraction group,the rats in the water extraction group showed low spirit,reduced physical activity and food consumption,and more serious mycardial tissue damage.Compared with model group,the serum levels of LDH,MDA and CK were decreased and the SOD activity was increased in the rats of the water extraction group and percolation extraction group;the serum level of BNP in the percolation extract group was obviously decreased,while that in water extract group did not.In both Shengmaiyin groups,the systolic pressure(SP),diastolic pressure(DP),arterial pressure(AP),and-dp/dtmax increased significantly compared with model group.Conclusion:Shengmaiyin prepared by percolation extraction method has better therapeutic effects on rats with chronic heart failure than the aqueous extract.
