一种快速检测小麦赤霉病菌种类的方法

为建立一种能够快速、简便和准确检测我国小麦赤霉病菌种类的方法,直接采用发病麦穗快速提取DNA,并结合特异性分子标记对采自我国的小麦赤霉病菌种类进行快速检测。该检测方法成功实现了对小麦赤霉病菌基因组DNA的快速提取和特异性分子标记检测,通过对河北、河南、江苏、安徽四省41份样本的检测验证,结果显示,安徽合肥与江苏南京地区的18份材料均为亚洲镰刀菌(Fusarium asiaticum),河北安新与赵县地区的12份材料均为禾谷镰刀菌(Fusarium graminearum),而河南地区的11份材料中2种致病菌株均有出现,检测结果与已经报道的这2种致病菌的地区分布情况一致。该方法不仅可以达到传统方法(CTAB法提取DNA)的效果,而且与之相比还具有快速、简便的优点,可用于小麦赤霉病菌群体结构和地区分布情况的研究。

A Rapid Detection Method for Gibberella saubinetii

To establish a rapid,simple and exact method for detection of Gibberella saubinetii in China,this study extracted DNA by using diseased ear directly,and combined with specific molecular marker for the rapid detection of Gibberella saubinetii.The genomic DNA of Gibberella saubinetii could be rapidly extracted and successfully detected by specific molecular marker with this detection method.Through the detection and verification of 41 samples from four provinces(Hebei,Henan,Jiangsu and Anhui provinces),the results showed that 18 samples from Anhui and Jiangsu were Fusarium asiaticum,12 samples from Hebei were Fusarium graminearum,and two pathogenic strains existed in the 11 samples from Henan.The detection result consisted with the situation of regional distribution of two pathogenic strains.The method can not only achieve the effect of traditional methods(DNA is extracted by CTAB method),but also have the advantages of rapidness and simplicity.It can be used in the study of population structure and regional distribution of Gibberella saubinetii.
