摘    要:目的:采用毛细管区带电泳法(Capillary Zone Electrophoresis,CZE),利用β-环糊精(β-cyclodextrin,β-CD)和羟丙基-β-环糊精(Hydroxypropyl-β-cyclodextrin,HP-β-CD)作为手性添加剂,拆分肾上腺素(Epinephrine)手性对映体,并检测市售盐酸肾上腺素注射液的手性状况。方法:通过对缓冲液体系、酸碱性以及β-CD和HP-β-CD的比较研究,得到优化的分离条件,即50mmol/L的Tris-H3PO4(用磷酸调pH为2.42),含40mmol/L的HP-β-CD,二极管阵列检测器检测,监视波长214nm,分离电压30kV,分离温度25℃,分离时间15min,压力进样5s。结果:肾上腺素((±)-Epinephrine)手性对映体在HP-β-CD体系中获得了完全的拆分,而β-CD对其没有拆分效果。检测发现市售的盐酸肾上腺素注射液为单一手性化合物。结论:毛细管区带电泳结合HP-β-CD手性添加剂可以有效的拆分肾上腺素手性对映体,并可作为检测产品纯度的有效方法。

Separation of epinephrine enantiomers using Capillary Zone Electrophoresis

Abstract:Objective:To establish a method based on Capillary Zone Electrophoresis to separate epinephrine enantiomers.Methods:The electrophoretic condition and parameters were investigated,and the optimized conditions were as follow ing:the electrophoretic medium was 50 mmol/L Tris-H3PO4 buffer(pH adjusted to 2.42 with H3PO4) containing 40 mmol/L HP-β-CD as chiral selector.Determination was carried out with detector of DAD at 214 nm.The separation were per formed at 25℃ with a positive voltage of 30 kV.Samples were injected into the capillary by pressure for 5 s.Results:The epinephrine enantiomers were separated entirely when they were in HP-β-CD buffer.Conclusion:CZE combined with HP-β-CD as an adjunct is a valid method to separate the epinephrine enantiomers.
