黄芪总黄酮对柯萨奇B3病毒感染心肌细胞动作电位的作用

摘    要:目的探讨黄芪总黄酮(TFA)对嗜心性柯萨奇B3病毒(CVB3m)感染心肌细胞动作电位的作用。方法采用体外培养小鼠新生心肌细胞方法,建立柯萨奇B3病毒感染心肌细胞模型,应用膜片钳技术中电流钳记录柯萨奇B3病毒感染心肌细胞动作电位。结果应用TFA组与柯萨奇B3病毒感染模型组相比较,动作电位幅值(APA)下降,差异有统计学意义(P=0.0065,n=8),动作电位时程(APD)明显延长(P=0.00724,n=8)。结论黄芪总黄酮可显著降低柯萨奇B3病毒感染心肌细胞动作电位幅值(APA),延长动作电位时程(APD)。

The effects of totalflavonoids of astragalus (TFA) on the action potential of the cardiomyocytes infected by coxsackievirus B3

Abstract:Objective To study the effect of totalflavonoids of astragalus (TFA) on the action potential of the cardiomyocytes infected by coxsackievirus B3. Methods A toxic celluar modell was estabished in vitro by adding CVB3m into the culture of neonatal mouse cardiomyocytes, the AP tracing of the cardiomyocytes infected by coxsackievirus B3 were recorded by whole cell membrane current clamp. Results TFA (20 mg/kg) reduce the action potential Amplitude (APA)(P=0.0065, n=8)and prolong the action potential duration (APD) of the cardiomyocytes infected by coxsackievirus B3 (P=0.00724,n=8). Conclusion The above results suggested that TFA reduce the action potential Amplitude (APA) and prolong the action potentiathe duration (APD) of the cardiomyocytes infected by coxsackievirus B3 remarkably.
