青龙衣中胡桃醌对A-549细胞周期及凋亡影响

摘    要:探讨青龙衣中胡桃醌(Juglone)对体外培养的人肺癌细胞A-549生长抑制作用及对细胞凋亡和周期的影响.采用MTT法检测青龙衣中胡桃醌对A-549细胞的生长抑制作用,采用激光共聚焦扫描显微镜观察凋亡形态,流式细胞仪分析胡桃醌对A-549细胞周期的影响.MTT法试验结果显示,胡桃醌对体外培养的人肺癌细胞A-549具有明显的生长抑制作用,处理24、48、72 h的IC50值为分别为3.4×10^-5、1.8×10^-5、2.6×10^-6mol/L.凋亡形态观察结果显示,胡桃醌能诱导A-549细胞的凋亡,同时胡桃醌能剂量依赖性的降低A-549细胞的G1期细胞的比例,升高G2期和S期细胞比例,呈G2期和S期阻滞作用.胡桃醌对A-549细胞具有明显的生长抑制作用,阻止周期进程,诱导A-549细胞凋亡.

Abstract:To explore the inhibition of juglone in Qinglongyi on A -549 cells in vitro, MTr assay is used. Laser confocal scanning microscope is used to observe apoptotic morphology. Changes of cell cycle are studied by flow cytometry analysis. The result of MTT assay showed that juglone had a marked growth inhibition in A -549 cells and the IC50 is respectively 3.4 ×10^-5 Stool/L, 1.8 ×10^-5mol/L and 2.6×10^-6 mol/L after treatment for 24, 48 and 72 h by juglone. Through laser confocal scanning microscope, it can be concluded that juglone can induce the apoptosis. Cell cycle changes are analyzed by flow cytometry with cells at G1 phase significantly less than those of control and cells at G2 phase significantly more than those of control. The result suggests that juglone could apoptosis of A - 549 cells with the cell cycle arrest on G2 and S phase in distinct dose-dependent manner.
